Study on the Influence of Various Suppository Bases on the Rectal Absorption of Pyrazinamide



Abstract
This paper attempts to investigate the influence of each suppesitory base on the rectal absorption of pyrazinamide and to compare the rectal administration with oral administration in blood level of pyrazinamide.
Rectal administration has no gastrointestinal disturbances and the nausea as a side-effect and also has very good velosity of absorpotion. For drugs are absorbed from the mucous membrane and transferred directly into the venous circulation. The results determined are as follows
1) It is found that among various suppository bases, hydrophillic bases in blood level of pyrazinamide show the highest point and emulsion bases show intermediate point and lipophillic bases show the low point.
2) When it comes to compare oral administration with rectal administration in blood level of pyrazinamide, hydrophillic bases are similar to oral, and emulsion bases are lower than oral and Hydrophillic bases, but among emulsion bases, lanett wax showes good results, and¢¥ lipophillic bases are far lower than oral.
3) Absorption peak point in oral administration is 120 minutes but absorption peak point in various suppository bases is largely 60?70 minutes, which is more rapidly than that of oral.
4) P.E.G., lanett-wax, and glyceronated-gelatin are similar to oral administration in blood level of pyazinamide, which are considered as a excellant suppositroy bases in therapeutic effects.
